I get emotional that at that point, David had hardly seen Hook interact with Emma but he can already sense a fundamental change in him. A glimmer of hope. Prior to this, a large part of Hook wanted to die. None of his enemies would grant him this. Then he met Emma. In their small amount of time together, his despair began to fall away. And David noticed it probably before Hook did.
